I am looking to create a website, the criterea ;
1.A secure Membership section with a password expiration of say 30 days, and automatic password renewal.
2 Configurable Membership application form.
3. Members able to download files up to 32Mb
4. Be compatable with Access database ( for listing file info)

Have you anything 'out of the box' that can give me this?

Items 1-3 are certainly part of DotNetNuke. The "Secure Membership Application" item might be satisfied by the DotNetNuke Registration page, or might need something more flexible such as the Form & List module. If you really need to get creative with site registration and membership, you can't go wrong by including DataSpring' Dynamic Registration module. And, depending on your exact application, you can combine it with their Dynamic Login and Dynamic Forms module. That's a real powerful combination.

With respect to Item 4, what exactly do you want to do with Access? DotNetNuke requires a Microsoft SQL Server and database. However, if you want DotNetNuke to also manipulate data from a separate Access database, that is possible with both "stock"( Form & List and Reports) modules as well as third-party modules. Depending, you'd probably be better off if you can convert your Access data to MSSQL tables.
