Can you provide any more details? Is this a new site? Has this started happening in an old site? If so, what did you change?

You'll need to determine if this is a DotNetNuke issue, and ASP.NET issue, or an IIS issue.

To check out IIS, but a file named index.htm in the web directory. Then browse to the site. If you see that index.htm is being served, then IIS is ok.

Next, but an file named app_offline.htm in that directory (after removing index.htm). Is that file served?

If both of those files are served, then you have something wrong with DotNetNuke. More details will help understand what it is.
