Have you tried to restart the website, or reboot the server?

When did this happen. Did you Make any changes to your server before this occurred or to any of the following files/things.

web.config

IIS

Security Permissions

Application Pool

It may just need a reboot as Joseph said but if that does not work have a look at what I said.

No one had accessed the site for a couple weeks. I rebooted the server and restarted the site and neither of those worked. We keep a daily backup up to 30 days prior so I went to a restore from a couple weeks ago and that worked fine. I have no idea what happened.

Well at least you had a backup to fall back on I believe the most likely explanation is that a file or the database has changed/corrupted in such a way it would cause this to happen. If you ever get this problem again please backup the site with the error and forward it to me as I am currently trying to map the DNN errors for one of my sites to advice people on quick fixes so they do not lose there changes in a single day or week. Because I have reloaded a backup once on a site I had and there were 40 new signups that were missing as it was the database that was damaged. A later found a fix but I had already restored the backup from the previous day.
