I need to set up my DNN site to accept registrations but I don't want the new users automatically given access.  I believe this is a function of the Auto Assignment check box.  My problem is that I can't uncheck that box because it it grayed out. 

I want to allow new users to register and then physically allow access if they "qualify".  Is Auto Assignment the correct place to do that?

Generally, the notion of a Registered User is that they have registered -- created an account -- on your site. You would set up another security role for subscribers. I would not auto assign that role.

I set up the register link and the form it brings up is fine, but it allows for people to become registered users.  How do I set up a link so that people can become subscribers and when they subscribe I can approve their subscriptions?  Is there a tutorial on this subject?

Normally, you would add "subscribers" to a suitable subscriber role manually.  Look at these tutorials for handling security roles and permissions:
