Can you mention the steps you followed to upgrade your application to 5.1?

I will assume that putting an index.html file in your web directory still is served correctly.

Rename the index.html file to app_offline.htm.  When you browse to your site, that file should be served.  (I like to make a small change to the file, so that I know that it really is the app_offline file that is served.)

If app_offline is served, that means that ASP.NET is installed correctly on your web server.  If it isn't, then you need to look at the ASP.NET installation.

Let me know what happens when you do that.

Thanks for ur valuable suggestion. Renamed the file as app_offline.htm i can able to see the page in my site. Report the details to server maintenance team. Actually its IIS problem they check it and resolved the problem. Now my site is Up. Thanks a lot. I will ask them what changes they made in IIS and i will post it in forum it will help others.

Thanks,
Anu

Can you mention the steps you followed to upgrade your application to 5.1?

This should help:
How to Upgrade DotNetNuke (Auto Merge Method) (v4.6.2 to v5+)

Its the same method for upgrading for all versions of 4.6.2 and above.

The most important thing you can do is backup, backup, backup.
